2. Description of the traits and characteristics which have been introduced or modified, including marker genes and previous modifications:
The GM-potato is characterized by a higher level of late blight resistance. Depending on the construct, the GM-potato contains the selection marker gene nptII or ahas, or is marker free.

In this project 10 different constructs are used which consist of different combinations of the following fragments:
- T-DNA borders from Agrobacterium tumefaciens for stable incorporation into the plant genome (constructs 1–10).
- The nptII gene from Escherichia coli, to be used as selection marker (constructs 1–5, 8, 9).
- The ahas gene from Arabidopsis, to be used as selection marker (construct 6).
- Nopaline synthase promoter from Agrobacterium tumefaciens for regulation of nptII or ahas expression (constructs 1–5, 6, 8, 9).
- Nopaline synthase terminator from Agrobacterium tumefaciens for regulation of nptII or ahas expression (constructs 1-5, 6, 8, 9).
- The Rpi-blb1 gene (including original promoter and terminator) from Solanum bulbocastanum for late blight resistance (constructs 1, 5, 6).
- The Rpi-blb2 gene (including original promoter and terminator) from Solanum bulbocastanum for late blight resistance (constructs 2, 6).
- The Rpi-blb3 gene (including original promoter and terminator) from Solanum bulbocastanum for late blight resistance (constructs 3, 8, 9, 10).
- The Rpi-R3a gene (including original promoter and terminator) from Solanum demissum for late blight resistance (constructs 4, 5, 7, 9, 10).
- The Rpi-sto1 gene (including original promoter and terminator) from Solanum stoloniferum for late blight resistance (constructs 8, 9, 10).

6. Brief description of the method used for the genetic modification:
T-DNA was introduced into potato cultivars by Agrobacterium-mediated gene transfer technology using a binary vector system.

Not applicable.

Experimental Release

1. Purpose of the release:
- Production of seed potatoes for future trials.
- Evaluation of late blight resistance under field conditions.
- Evaluation of different measurements for late blight control.
- Evaluation of the agricultural value of modified genotypes.

2. Geographical location of the site:
Municipalities: Borger-Odoorn, Eemsmond, Coevorden, Rhenen, and Wageningen. All municipalities are situated in The Netherlands.

3. Size of the site (m2):
The size per site will be variable. The maximum size per site is 10,000 m2

4. Relevant data regarding previous releases carried out with the same GM-plant, if any, specifically related to the potential environmental and human health impacts from the release:
This is the first release of these GM-plants. Cultivars, derived by classical breeding methods, containing the described and/or comparable late blight resistance genes are being cultivated for several decades without any negative impact on the environment or human health.

Environmental Impact and Risk Management

Summary of the potential environmental impact from the release of the GMPts:
The introduced resistance will not confer an increased selective advantage in natural environments compared to the effect of late blight resistance genes present in potato cultivars.

Brief description of any measures taken for the management of risks:
All handling of GM-plants and GMO fields will be done according to good agricultural practices.
- Transportation of potatoes will be done in bags in open containers.
- Crop rotation: no potato crop the following two years.
- Destruction of volunteers
- Destruction of waste material.
